FAQs - Metabolomics
1. What is metabolome?
Metabolites represent an extremely diverse range of molecules including amino acids, lipids, hormones, carbohydrates, etc. The compilation of endogenous metabolites within a biological system is comprehensively known as the "metabolome".
2. What is metabolomics?
Identification and quantification of metabolites (small molecule less than 1500 Da) from “metabolome” is called metabolomics.
3. What are the ways to perform metabolomics experiment?
It can be done in both targeted (you should know which molecules to be quantified) and untargeted (trying to analyse all the molecules in a given sample) analysis.
4. What kind of instruments will be suitable to do these experiments?
For profiling one has to use the high resolution instruments (QTOF, Orbi-traps) and for quantification (Triple Quad or Qtrap) instruments.
5. What is the difference between metabolite profiling and metabolite quantification?
In profiling you are trying to analyse all possible molecules in the given sample using high resolution instruments and in the quantification you are interested only in certain compounds.
6. What kind of service that you are providing right now?
We are providing only the targeted metabolite quantification using LC-MS. Look into our protocols for more details. We have an established method to quantify neurotransmitters from biological fluids.
7.How do you quantify metabolites using isotopically labeled internal standards?
Normally this involves the use of a stable isotope-labeled internal standard, which is spiked into a sample at a known concentration. The response ratio between the analyte and labeled compound obtained by LC–MRM-MS can then be interpolated onto a standard curve to calculate the absolute amount of analyte in the unknown sample.
8. What is absolute quantification?
By using specific internal standards (ISTDs) trying to quantify the molecule of interest with the help of advanced LC-MS system.
9. What kind of control biological matrices you have?
We have double charcoal stripped human sera which can be used as a control matrix for sera and plasma to develop the method. For any other matrices either you have to provide the control matrix or inform in advance so that we can procure it from the company.
10. Do I have to provide the standard and internal standard to standardize the method?
It will be good if you could send us both the standard and internal standard otherwise you can tell in advance so that we will procure it from the company (you have to pay for it) to develop the method.
11. What kind of sample to be submitted for the quantitative analysis?
Any kind of biological matrices (sera, plasma, saliva, tissue, urine, plant extract and cell extract). If you have an established protocol to extract metabolites you can process it and give us the dried product.
12. How much sample needed?
For a typical full scan experiment we need (1-10 ug/mL sample). The full scan experiments will be performed only for the known/purified metabolites. For targeted experiment we need (ng to pg range).
13. What is the turnaround time for a service?
Normal full scan experiment will take 1-2 days. Quantification will take 1-2 weeks for the established method and 6-8 weeks for the new method followed by analysis.
14. What are the fees for the services?
We follow different fee structures for internal academia, external academia and commercial users. The cost may also differ depending on particular research question you ask us to solve for you. For further details please write to Mr. Mohan Sridhar at mohans at ncbs dot res dot in.
15. How will you deliver my data? Can I get my raw data files?
The data on mass measurement will be sent as PDF slides/document and validation data will be sent as Excel sheets. About raw data, we prefer that data is not given out of our hands. If you feel it is very necessary under special circumstances, kindly get back to us and we will guide you through our policies of raw data sharing.
16. Should I acknowledge the facility in my publications and presentations?
It is mandatory to acknowledge the MS-Facility in your achievements. In case of companies or organization which provides the data to the third party, please check our website at

for our Raw Data and Data Storage policy. It is a must to also formally acknowledge the facility in writing in the case of providing our report to a third party and also declare us as a preferred MS partner of your company/ organization.
17. What is a work order?
A work order is request made from your Institute/Organizations Purchase Department for the service we are providing. It needs to be made against the quotation that we provide.
